Gossip website Holy Moly initially broke the story earlier this month that the 'Crazy in Love' star would close proceedings on the Sunday night.
Now, the singer has confirmed the reports herself, telling Billboard: "This really is the biggest festival in the world and I cannot wait to perform there.
"Everyone who attends is really appreciative of music and is in such a good mood that entire weekend. I'm pumped just thinking about that huge audience and soaking up their energy."
The Libertines have also been confirmed for the festival, following their reformation for last year's Reading Leeds festivals.
It's thought that U2 will headlining the first night, with Coldplay already confirmed to headline the Saturday.
Beyonce visited the festival in 2008, when her husband Jay-Z controversially headlined, but she has never performed at the event.
Glastonbury 2011 takes place at Worthy Farm near Pilton in Somerset from June 24 to 26.
Last year Gorillaz replaced U2 as headliners when Bono had to pull out with a bad back, with Muse and Stevie Wonder also topping the bill.
