For Thom Yorke and guitarist Jonny Greenwood, it will be their second consecutive year playing the stage after the duo appeared as special guests 12 months ago.
Friday night's show will be the full line-up's fourth Glastonbury outing after first appearing in 1994, before returning three years later for what many consider to be the finest display Glastonbury has ever seen.
It's long been a tradition for rumours of secret sets to ripple around the massive Worthy Farm site, however this year it would appear that not even organiser Emily Eavis is entirely sure just who will be playing.
She told the NME, "We are still unsure of exactly who is turning up, mainly because it's unconfirmed at the moment! I swear I have heard every rumour -- even that Led Zeppelin are playing!"
It's not just fans who continue to speculate over who else might be added to the bill. Two Door Cinema Club told Gigwise, "We've heard a lot of talk. Pulp, Arctic Monkeys, Prince, Jay-Z might do one. We've heard Arcade Fire as well."
